Murine hemopoietic stem cell proliferation and differentiation are controlled by a delicate balance of hematopoietic growth factors and their cognate cellular receptors. Receptor-negative thymic stromal cells have been recently shown to produce several cytokines with hemopoietic growth factor-like activity including T cell growth factor/interleukin 2 (TCGF/IL2), macrophage-CSF/interleukin 4 (M-CSF/IL4), erythropoietin (Epo), and interleukin 7 (IL7). To determine if this cytokine network supports in vitro hematopoietic development, we have cultured thymic organ cultures from adult BALB/c mice in the presence of recombinant cytokines. With the exception of IL7, all cytokines supported thymocyte growth and increased the proportion of CD4-8- cells. Cytokine combinations required for optimal hematopoietic growth (IL7 plus M-CSF/IL4 or Epo plus M-CSF/IL4) demonstrated either nonoverlapping or synergistic growth effects. In contrast, the presence of IL7 and M-CSF/IL4 promoted increased CD4+/CD8+ T cell frequency and augmented T cell receptor beta gene rearrangement. Macrophage-derived growth factors (IL4 or M-CSF) did not reverse the growth inhibition caused by IL7. IL7 promoted increased nuclear factor of activated T cell (NFAT) activity and synergized with a calcium ionophore, thereby inducing T cell receptor beta gene rearrangement. Taken together, these results suggest that the thymic microenvironment contains a network of cytokines which supports thymocyte differentiation and growth.
